O-Level Biology Leaf and Stem Microscopy: Identify Tissues Safely
Observe prepared leaf and stem sections, recognise their tissues and make biological drawings while handling slides and focusing the microscope carefully.
A leaf section shows how tissues are arranged through the leaf. A stem section shows the position of its vascular tissues. Begin with the whole section before trying to name individual cells: their position is part of the evidence.
This guide uses prepared transverse sections of a typical dicot leaf and a young dicot stem. It does not require students to cut tissue or add stains. Follow the teacher's specimen instructions and the microscope's operating procedure. For the wider preparation habits, read Lab safety starts before the experiment.
What the syllabus asks you to recognise
The 2026 SEAB Biology syllabus, 6093, page 18, includes identifying dicot leaf structures and explaining their functions, and identifying xylem and phloem in leaf and stem sections under a light microscope. Observing an epidermal peel for stomata is a different view from observing a transverse section through the leaf.
The prepared-slide example supports observation and identification. It is not a claim that every student must prepare fresh sections, or that these are the particular slides used in an Eclat lesson.
Find the specimen before increasing magnification
Check the slide label and its edges before placing it on the stage. Report cracked or chipped glass and ask for a replacement. Hold slides by their edges without pressing on the thin coverslip; return unused slides to their tray rather than stacking them on the bench edge.
Start with the lowest-power objective. Secure the slide gently using the stage arrangement provided. Looking from the side helps you check the clearance between the objective and the slide before focusing. Locate the tissue, bring it into focus and centre the region you want to inspect.
